What’s the difference between HPLC purity and endotoxin levels?

HPLC (High-Performance Liquid Chromatography) measures the purity of a peptide sample – what percentage of the detected material is the target peptide versus impurities or byproducts. A 98%+ HPLC result means the peptide content is highly concentrated.

Endotoxin screening (typically LAL testing) measures bacterial endotoxin contamination – a completely different parameter. Endotoxins are bacterial cell wall fragments that can be present even in a highly pure peptide. Results are reported in EU/mL or EU/mg (Endotoxin Units), where lower numbers indicate lower contamination.

Why both matter:

  • HPLC purity tells you what’s in the vial
  • Endotoxin screening tells you what else might be in the vial
  • A peptide can pass HPLC at 99% purity and still have elevated endotoxins – or vice versa

How can I verify a product’s Certificate of Analysis (COA)?

Why does my vial look almost empty?

Research peptides are supplied in lyophilized (freeze-dried) form to improve stability during storage and shipping. The freeze-drying process removes moisture, which means the peptide may appear as a small white puck, fluffy powder, thin film, or light residue at the bottom of the vial — even when filled correctly.

This is normal and not an indication of underfill. The label weight (e.g., 5 mg, 10 mg, 30 mg) refers to the mass of the peptide itself, not the physical volume of powder. Some peptides are inherently lightweight and fluffy, so a visually small amount can still be precisely the correct dose.

For laboratory research, lyophilized peptides are reconstituted by adding an appropriate solvent per the researcher’s protocol. Vials should be brought to room temperature while sealed before reconstitution to prevent condensation inside the vial.
